To meet the need of labeling the broadest range of target molecules each fluorescent compound is available with many different reactive moieties (e.g. the commonly used amine reactive and thiol reactive groups) while retaining the photophysical features of the parent dye. Furthermore, a new level of chemistry has been developed to provide the same fluorescent label with different charge values (anionic, cationic, zwitterionic and neutral). It is thus easy to find the ideal product among the Colorada series starting from the desired absorption/emission peak maximum and subsequently decide both the reactivity and charge properties. Different basic structures with precise absorption and emission maxima were selected to cover the full working range of wavelength, starting from 400 nm up to 850 nm: Coumarin dyes, Dipyrrometheneboron difluorides (BDPs), Trimethine cyanine dyes, 8-Oxo-8H-cyclopenta[a]acenaphthylene-7-carbonitriles (Acenaphthopyrrole dyes), Pentamethine cyanine dyes, Heptamethine indocyanine dyes which includes the dyes with a Large Stokes Shift (Colorada 670 LSS and Colorada 690 LSS).
The reactive groups include carboxylic acid (-COOH), active ester (-COOX, NHS ester), dichlorotriazine, aliphatic amine (-NH2), azide (-N3), iodoacetamide and 2-pyridyldisulfideamine (PDA), the latter being very useful for labeling -SH in peptides or proteins. Colorada Antibody Labeling Kits
The Colorada Antibody Labeling Kits were developed for a fast and highly efficient protein labeling procedure. They are specifically designed for the fluorescent labeling of 1 mg of antibody, typically IgG. An extraordinary feature of the kits is the ease of handling: The reaction is carried out by simply mixing the supplied reagents and adding the target antibody. No weighing of tiny amounts of expensive and unstable activated dye is needed. By keeping the kit reagents separate, maximum stability of the dye is achieved. Volumes and amounts are carefully tuned in order to obtain maximum fluorescence and preserve antibody activity, with an optimal 5-10 dye-to-protein ratio. The two-vial formulation has the additional advantage of generating the reactive dye quantitatively in situ, thus providing maximum efficiency in the conjugation step.
Colorada DIGE Labeling Kits
The Colorada DIGE Labeling Kits are specifically designed for the detection of proteins in 2-D Fluorescence Difference Gel Electrophoresis (DIGE). The kits are supplied with either Colorada 490, 550 or 678 DIGE Dye, respectively, highly fluorescent fluorochromes, based on the cyanine structure. The minimal labeling approach, the basic idea of these kits, requires a minimized concentration of the dye in order to label each protein with a single dye molecule. Labeling occurs by forming an amide bond between the ε-amino group of lysine in proteins and the activated dye. The in-situ activation of the dye affords maximum efficiency in the labeling step along with longer stability of the product. The special design of the Colorada's results in:
● Charge-matched: A positive charge on each dye replaces the one lost from lysine upon formation of the amide bond, leaving the total charge on the protein unchanged.
● Size-matched: All dyes have a similar molecular weight to minimize differences in protein migration after labeling.
● pH insensitive: The fluorescent signal is not influenced by the pH value over a wide pH range.
